The V-12 basic curriculum was organized into two 16-week terms. Courses of instruction included mathematics, chemistry, English, physics, engineering, drawing, physical training, naval organization and the historical background of World War II. Each Sailor was expected to attend an average of thirty-two class hours of instruction each week.

U.S. Navy sailors and civilian students were integrated at MSTC. As a majority of the instruction the Sailors received at MSTC was in general education, which was required for all students, civilian students pursuing degrees attended classes side by side with Navy personnel.

Upon completion of two terms at MSTC, Sailors transferred to Midshipman's School or other V-12 colleges for additional training, in either general or specialized skills. While the Sailors were not granted a degree directly from MSTC, the credits they received did apply to a degree that was awarded upon completion of the full V-12 program.
